Core Responsibilities

Primary Functions

  • Perform a variety of standard and advanced molecular biology techniques, including DNA/RNA extraction, PCR, qPCR, and RT-PCR, to analyze vaccine components and host responses.
  • Execute a diverse range of immunological assays, such as ELISA, ELISpot, and multiplex bead assays (e.g., Luminex), to quantify antibody titers and cytokine profiles from pre-clinical or clinical samples.
  • Maintain and passage mammalian cell lines using sterile, aseptic techniques, ensuring cell health and viability for use in viral infectivity, neutralization, and other cell-based assays.
  • Conduct viral plaque assays, TCID50 assays, and neutralization assays to determine the potency and efficacy of vaccine candidates against viral challenges.
  • Prepare and formulate vaccine candidates, adjuvants, and other critical reagents with high precision, following detailed standard operating procedures (SOPs).
  • Operate and maintain sophisticated laboratory instruments, including flow cytometers, plate readers, automated liquid handlers, and microscopes, performing routine calibration and troubleshooting.
  • Process and analyze biological samples (e.g., blood, serum, plasma, tissues) from in-vivo studies, preparing them for downstream analysis with careful attention to sample integrity.
  • Assist in the design and execution of experiments under the guidance of a supervising scientist, providing input on experimental setup and potential challenges.
  • Meticulously document all experimental procedures, raw data, and observations in electronic lab notebooks (ELNs) in a timely and compliant manner.
  • Analyze experimental data using software such as GraphPad Prism, FlowJo, or Microsoft Excel, and prepare preliminary summaries, figures, and graphs for review.
  • Support in-vivo studies by assisting with animal handling, dosing, and sample collection, strictly adhering to all IACUC and animal welfare guidelines.
  • Participate in the development, optimization, and validation of new assays and protocols to enhance the lab's research capabilities.
  • Culture and isolate primary immune cells from blood or tissue and utilize them for functional assays that measure T-cell and B-cell responses to vaccination.
  • Perform multiparameter flow cytometry staining and acquisition to characterize and phenotype various immune cell populations.
  • Prepare detailed technical reports, summaries, and presentations of experimental results for internal lab meetings and project discussions.
  • Contribute to a robust and safe lab environment by strictly following all BSL-2 (or higher) safety protocols and procedures for handling infectious agents.
  • Troubleshoot experimental workflows and assays, identifying deviations from expected outcomes and working with senior staff to implement corrective actions.
  • Manage the cryopreservation of cell banks, viral stocks, and valuable biological samples, maintaining an organized and accurate inventory system.
  • Collaborate effectively with cross-functional team members, including virologists, immunologists, and process development scientists, to advance project goals.
  • Stay current with relevant scientific literature and new technologies to contribute fresh ideas and improve laboratory methods.

Secondary Functions

  • Maintain detailed and accurate records of all experiments and results in compliant electronic lab notebooks (ELNs).
  • Manage the inventory of laboratory reagents, consumables, and supplies, initiating purchase orders to prevent work disruptions.
  • Assist in the general upkeep, organization, and maintenance of the laboratory space and common equipment.
  • Actively participate in weekly lab meetings and scientific discussions, presenting experimental findings and contributing to project planning.

Required Skills & Competencies

Hard Skills (Technical)

  • Aseptic Cell Culture: Proficiency in maintaining and manipulating mammalian cell lines in a sterile environment.
  • Immunological Assays: Hands-on experience with ELISA, ELISpot, and/or multiplex platforms (Luminex).
  • Molecular Biology Techniques: Competency in nucleic acid extraction, PCR, and qPCR.
